There are many advantages to these systems, yet they also have significant limitations. cell cultures offer the opportunity to investigate the cell basics or primary response of a cell population against any treatment. However, these models do not always predict behavior. animal studies constitute the most realistic platform for research and therapeutic approaches, yet they are laborious, open to secondary complications and painful or stressful for the animals from an ethical point of view. Organotypic brain slice cultures provide an -like environment since they maintain three-dimensional cytoarchitecture of the brain thus enable to study many cell types in one system and allow precise control of the microenvironment. In this review, we will focus on the history and key features of organotypic brain slice cultures as well as its preparation.
