A fermentation system was designed that utilizes
n-paraffins as the main carbon/energy (C/E) source for penicillin biosynthesis, based upon the oxidation of phenyl-n-alkanes to phenyl acetic acid by
Penicillium chrysogenum.
n-Paraffins (component A) were found to be a suitable replacement for carbohydrate in penicillin fermentations when combined with an easily metabolizable substrate, triglycerol ester (component B). Some physico-chemical parameters (surface and interfacial tensions) of the oil mixture were determined to calculate the spreading force (
S), a contributing factor for adequate consumption rates of the non-aqueous liquid phase. Using a fed-batch-type fermentation system, key metabolic indicators, such as specific growth rate, residual
n-paraffins,
RO
2 and RCO
2, with its corresponding
RQ, are presented. The applied technique in the fermentation conduction supports a penicillin synthesis rate comparable to the control.
