  • Liquid biopsy is an emerging technology for disease diagnosis and monitoring, which can detect and analyze tumor‐derived circulating targets in body fluids, including circulating tumor cells (CTCs), extracellular vesicles (EVs), proteins, circulating free DNA, and microRNAs. This chapter summarizes the recent advancements in DNA nanotechnology‐based microfluidics for liquid biopsy. It also summarizes recent progress in DNA nanotechnology‐based microfluidics for the isolation of circulating targets. The chapter describes the use of DNA nanotechnology‐based microfluidics for the release and detection of circulating targets. It discusses the applications of DNA‐assisted microfluidics for single cell/vesicle analysis. The chapter provides future perspectives and challenges in the field of DNA nanotechnology‐based microfluidics for liquid biopsy. The powerful encoding capability of DNA, combined with second‐generation sequencing, can facilitate high‐throughput parallel analyses of targets within blood circulation, especially CTCs and EVs. The miniaturization feature of microfluidics increases reaction kinetics in picoliter‐to‐nanoliter reaction system for rapid and sensitive detection.
