  • Curcumin from the curry spice turmeric is a polypharmacological agent with more than 100 reported cellular targets. Curcumin undergoes extensive metabolism in vivo and is also chemically unstable in vitro. Thus, discrepancies between metabolite formation in vivo and in vitro may contribute to differences in biological outcomes. The chemical instability of curcumin is well recognized but also widely misunderstood regarding the identity of the degradation products. Widely accepted is the notion that degradation of curcumin yields cleavage products like vanillin, ferulic acid and feruloylmethane. In contrast, recent studies using radiolabelled curcumin and product identification by LC‐MS and NMR show that the cleavage reaction is negligible and that degradation is an autoxidation that forms a double oxygenated bicyclopentadione as the major product. Autoxidation proceeds through intermediates that feature reactive electrophilic (quinone methide and epoxide) functional groups. Knowledge of the mechanisms of degradation of curcumin will enable a better understanding of how reactive intermediates and their formation in vitro and in vivo may contribute to the biological effects of curcumin.
