  • The Zona Pellucida (ZP) of all mammalian eggs, from platypus to human, consists of either three or four ZPD proteins, called ZP1–4. This chapter discusses aa sequences of ZP1–4 polypeptides for 12 mammalian species, from platypus (monotremes), to possum (marsupials), to human (placental mammals). Monotremes (Prototheria) are a mammalian sub‐class that includes the platypus and four species of echidnas (spiny anteaters) that lay eggs instead of giving birth to live young. Marsupials are represented by kangaroos, koalas, possums, opossums, wombats, and other less well‐known species. Placental mammals emerged as the dominant large animals on Earth after the extinction of the non‐avian dinosaurs more than 65 million years ago and today are represented by more than 5000 species. ZP proteins have proved very useful as immunogens for immunocontraception trials. Immunization with native or recombinant ZP proteins reduces fertility but is often associated with adverse ovarian pathology, long‐term infertility, or permanent sterility.
