From Molecular Science to the Treatment of Allergy First discovered over 60 years ago, H1-antihistamines have developed a fundamental role for management of allergic disorders. The second-generation drugs offer advantages of greater potency and reduced side effects. Simons et al. [1] compared these drugs and showed the order of potency for blocking the wheal and flare response to histamine in the skin was cetirizine, terfenadine, loratadine, astemizole, and chlorpheniramine. Juhlin [2] reviewed multiple clinical trials showing superiority for cetirizine. Recently, new H1-antagonists have become available, but there has been no comparison of their potency.
