Fibrin powder was found to aggregate or clump in the presence of fibrinogen. Aggregates not only scattered less light than the initial suspension of fibrin particles, but also sedimented more rapidly in tubes. Thus absorbance measurements after an appropriate settling time gave a more accurate estimate of the clumping effect than either initial absorbance or sedimentation rate measurements alone. Aggregation was found to be directly related to the fibrinogen concentrations in diluted plasma samples. It was not affected by heparin or other anticoagulants, but it was reduced by high levels of fibrin degradation products and by heat treatment of the fibrin powder. Measurements of fibrinolysis in plasma using fibrin powder substrate may be carried out using defibrinated samples and heat -treated fibrin powder suspension.
