The authors have attempted to induce an adaptive response in human fibroblasts by multiple treatments with N-methyl-N'-nitro-N-nitrosoguanidine (MNNG). The ability of treated cells to withstand a challenge by N-methyl-N-nitrosourea (MNU)was determined by cell survival (colony forming ability) and the induction of mironuclei. In addition, the authors have assayed two enzymes which are involved in the repair of methylation damage to DNA: O super(6)-methylguanine methyltransferase and 3-methyladenine-DNA glycosylase.
