  • Abstract Objective To construct a neonatal rat model of increased right ventricular afterload for studying the pathophysiological remodeling of increased right ventricle in congenital heart disease patients with right ventricular afterload. Methods Surgery was performed within 6 hours after birth. Horizontal thoracotomy was performed through dissecting intercostal muscles and splitting the sternum. Pulmonary artery was banded by an 11-0 nylon thread. At postnatal day 7, the constriction of pulmonary artery was confirmed by echocardiography. The right ventricular systolic and diastolic pressures were measured by cardiac catheterization. The right ventricular end-systolic volume, end-diastolic volume, end-diastolic diameter and free wall thickness were assessed by magnetic resonance imaging. The histological changes of sham and pulmonary artery banding hearts were evaluated after Hematoxylin and Eosin staining. Results Increased right ventricular afterload was established by constriction of the pulmonary artery in neonatal rats within 6 hours after birth. The survival rate is 75% at P7. Relative to the sham group, peak pressure gradient across pulmonary artery constriction, right ventricular systolic and diastolic pressures, right ventricular end-systolic volume, end-diastolic volume, end-diastolic diameter and free wall thickness were significantly increased in the pulmonary artery banding group at postnatal day 7 (P<0.01). Consistently, histological examination showed that the right ventricular free wall was significantly hypertrophic in the pulmonary artery banding group. Conclusions We successfully established an neonatal right ventricular afterload increase model through banding pulmonary artery within 6 hours after birth, which can be used to study the pathophysiological changes of congenital heart disease with increase right ventricular afterload.
