Using the recently developed technology known as antigen-reactive T-cell enrichment for a comprehensive characterization of human Treg cell responses, Bacher et al (Cell 2016;167:1067-78, e16; https://doi.org/10.1016/j.cell.2016.09.050) demonstrated dominant Treg cell-mediated tolerance against the aeroallergens pollen, house dust mites, and fungal spores in human subjects. [...]the loss of these 3 signals abrogated terminal differentiation of effector TH2 cells and adaptive lung inflammation in a T cell-intrinsic manner without affecting lymph node priming, suggesting that both ILC2s and TH2 cells are subject to a lymphocyte-intrinsic checkpoint that restricts type 2 cytokines to sites of tissue distress.
