Background
Canine atopic dermatitis (CAD) is a chronic dermatological disease partly due to dysregulation of the immune system. Inappropriate activation of CD4+ lymphocytes could favour and promote the allergic response. An inadequate activation system of regulatory T cells (Tregs) is suspected to be a key immunological feature of the allergic response in atopic dogs.
Hypothesis/Objectives
To evaluate the difference in the CD4/CD8 lymphocyte ratio and the percentage of Tregs in healthy dogs, in a breed predisposed to CAD, and in dogs affected by CAD before and during therapy with ciclosporin (CsA). Additionally to assess the improvement in pruritus and skin lesions during therapy with CsA, and to compare this with CD4/CD8/Treg values.
Animals
Ten atopic dogs of different breed, sex and age, ten healthy dogs and ten English bulldogs were included.
Methods
Peripheral blood from all dogs was tested using flow cytometry to assess the CD4/CD8 ratio and percentage of Tregs. For atopic dogs, sampling was repeated after 30 and 90 days of therapy with CsA.
Results
The CD4/CD8 ratio was not significantly different between the three groups. The Treg percentage was higher, but not statistically significant, in atopic dogs compared with controls. Therapy with CsA led to clinical improvement; it was not associated with statistically significant differences in haematological variables.
Conclusion and clinical importance
This study suggests that Tregs may be involved in the pathogenesis of CAD and that ciclosporin therapy does not affect the circulating lymphocyte subpopulations

Déterminer la différence du ratio des lymphocytes CD4/CD8 et le pourcentage de Tregs chez les chiens sains dans une race prédisposée à la CAD et chez les chiens atteints de CAD avant et pendant un traitement à la ciclosporine (CsA). Sujets
Dix chiens atopiques de différentes races, genre et âge, dix chiens sains et dix bouledogues anglais ont été inclus.
Méthodes
Du sang périphérique de tous les chiens a été testé par cytométrie de flux pour déterminer le ratio CD4/CD8 et le pourcentage de Tregs. Pour les chiens atopiques, des échantillons ont été répétés après 30 et 90 jours de traitement par la CsA.
Résultats
Le ratio CD4/CD8 n'était pas significativement différent entre les trois groupes. Hipótesis/Objetivos
evaluar la diferencia en el cociente de linfocitos CD4/CD8 y el porcentaje de linfocitos Tregs en perros sanos de una raza predispuesta a presentar CAD, y en perros afectados por CAD antes y durante la terapia con ciclosporina (CsA). Además se evaluó la mejora del prurito y las lesiones de la piel durante la terapia con ciclosporina, y se comparó con los valores de CD4/CD8/Tregs.
Animales
10 perros atópicos diferentes razas, sexo y edad, diez perros sanos, y 10 perros de raza bulldog inglesa se incluyeron el estudio.
Métodos

Hypothese/Ziele
Eine Erhebung der Unterschiede zwischen dem Verhältnis der CD4/CD8 Lymphozyten und dem Prozentsatz der Tregs gesunder Hunde, bei einer Rasse, die zu CAD prädisponiert ist, und bei Hunden mit CAD vor und während der Behandlung mit Ciclosporin (CsA). Zusätzlich sollte die Verbesserung des Juckreizes und der Hautveränderungen während der Therapie mit CsA erhoben werden und diese mit CD4/CD8/Tregs Werten verglichen werden.
Tiere
Zehn atopische Hunde unterschiedlicher Rasse, Geschlecht und Alter, zehn gesunde Hunde und zehn englische Bulldoggen wurden inkludiert.
Methoden
Peripheres Blut wurde von allen Hunden mittels Flowzytometrie getestet um das Verhältnis CD4/CD8 und den Prozentsatz der Tregs zu erheben. Bei den atopischen Hunden wurden die Proben am Tag 30 und 90 der Therapie mit CsA wiederholt.
Ergebnisse
Das Verhältnis der CD4/CD8 Zellen war zwischen den drei Gruppen nicht signifikant verschieden. Der Prozentsatz der Tregs war bei den atopischen Hunden im Vergleich zu den Kontrollhunden höher, wobei der Unterschied aber nicht statistisch signifikant war. Die Behandlung mit CsA führte zu einer klinischen Verbesserung; es konnte aber nicht mit statistisch signifikanten Unterschieden bei den hämatologischen Parametern in Verbindung gebracht werden.
Schlussfolgerungen und klinische Bedeutung
