A genomic DNA library of Arabidopsis thaliana was screened with a 2.2 kb XhoI-ClaI fragment of genomic DNA from Saccharomyces cerevisiae initially characterized as containing HEM15, the gene encoding ferrochelatase. DNA sequence analysis of the yeast DNA, reported in another communication, identified downstream to HEM15 an oppositely oriented tRNA super(val) (GUU) coding sequence. An A. thaliana DNA clone hybridizing with the S. cerevisiae probe was found not to contain a HEM15 homolog but a tRNA super(val) (GUU) sequence which can be folded into a cloverleaf structure. The A. thaliana tRNA is 75.6% similar to the yeast tRNA super(val) and included two mismatches and two insertions compared with the only sequence of a cytoplasmic tRNA super(val) from higher plants (from Lupinus luteus ) reported to date.
