  • A photomicrographic record is presented to show the morphology and behavior of centrioles in spermatogenesis of the hissing cockroach. In dissociated, air-dried, and silver-stained preparations, the centrioles of all stages of spermatocytes are clearly visible with bright-fleld microscopy. In very early melotic prophase, two pairs of centrioles (each with the mother and the daughter) lie side-by-side in the shape of two V's. The centrioles grow to the mature length in pachytene and the two pairs then migrate apart until they reach the poles at diakinesis. The mother and the daughter centrioles separate during first telophase and the cells of the second melosis have only two rod-shaped centrioles instead of V's, indicating no centriolar replication between the two meiotic devisions. After the second anaphase, the rod-shaped centriole moves away from the pole and attaches its proximal end to the spermatid nucleus and subsequently grows to become the sperm tall or flagellum while the polar end determines the position of the acrosome. Injection of ethidium bromide induces a variety of centriolar abnormalities. This system should be useful for analysis of the effects of chemicals on centrioles. The insects are large enough to withstand injections of a small amount of solution or suspensions into their abdominal cavities.
