Suitability of the pH Buffers 3-[N-N-bis(hydroxyethyl)amino]-2-hydroxypropanesul- fonic Acid and N-2-hydroxyethylpiperazine-N'-2-ethanesulfonic Acid for in Vitro Copper Toxicity Studies

In vitro copper toxicity studies were conducted using the marine dinoflagellate, Amphidinium carterae, to examine the impact of two pH buffers-3-[N-N-bis(hydroxyethyl)amino]-2-hydrox- ypropanesulfon- ic acid (DIPSO) and N-2-hydroxyethylpiperazine-N'-2-ethanesulfonic acid (HEPES)-on Cu toxicity. The experimental protocol is detailed. Results indicated that DIPSO was a strong Cu-complexing agent, which was not observed for HEPES. Neither buffer, at DIPSO and HEPES concentrations of 10 and 25 mM, respectively, was toxic to A. carterae. However, HEPES did not complex bivalent Cu significantly, which increased the toxicity of the metal, possibly due to changes in the kinetics of metal binding to the organism.
