  • The effect of DDT and three analogs on binding of 1-anilino-8-naphthalene sulfonate (ANS) by nonenergized mitochondrial and submitochondrial membranes, by delipidated submitochondrial membranes, and by liposomes prepared from submitochondrial lipids was examined. DDT enhanced the fluorescence intensity of the ANS-membrane complex. A graphical analysis of this effect suggests it to be due to an increase in the amount of ANS bound to the membrane and not to an increase in the quantum yield of ANS. The strongest enhancement of ANS binding was incurred by diphenylethane while the least was by DDT. For all compounds, the Scatchard plots from which the binding parameters, n and K d , were calculated, were linear. DDT did not enhance the ANS-binding capacity of delipidated submitochondrial membranes. A strong DDT-induced enhancement of ANS binding was, however, evident for liposomes prepared from the lipids of the membrane. These data suggest that the bulk of the interaction between DDT and the mitochondrial membrane occurred in the lipid phase. The possible mechanism of this interaction and the manner in which it accounts for increased binding of ANS is discussed.
