Various concentrations of semipurified cucurbitacin solutions were applied to chromatography papers. A colony of Diabrotica balteata , LeConte was allowed to feed on the treated papers, and visitations and feeding responses were recorded. Feeding response was reflected by the darkness of brown stains produced by insect saliva. Visitation frequency was a good indicator of the age of the colony; the parameter was also useful to estimate amounts of cucurbitacin, provided the colony was young. Feeding response declined slightly with increasing age. Dose-response curves showed that concentrations of cucurbitacins could be estimated fairly accurately within the bounds described (4-0.0078 mu g multiplied by mu l super(-1)). The threshold level was 5 x 10 super(-12) g, when applied in a volume of 0.2 mu l. Preliminary results indicated that the bioassay can be used to test for the presence of phagodeterrents.
