The demonstration of 1,25-(OH) sub(2)D sub(3) receptors (1,25-DR) in human breast cancer cells in culture, the uptake by intact cells and nuclear localisation, and the effects of 1,25-(OH) sub(2)D sub(3) on the replication of these cells in culture has provided us with a valuable in vitro model system for the examination of early biochemical events in the action of 1,25-(OH) sub(2)D sub(3) on target cells. The authors have used this model to examine the fate of hormone-receptor complexes once they have been specifically bound in the nucleus of the T47 D breast cancer cell. During these studies it was recognised that after maximum binding was reached there was a rapid decline in cell-associated radioactivity even in the presence of excess labelled 1,25-(OH) sub(2)D sub(3). This decline in specific binding implied a disruption of 1,25-(OH) sub(2)D sub(3) receptors complexes, which could be explained by inactivation of receptor, metabolims of hormone or a combination of both of these mechanisms. Experiments were performed to examine these cellular mechanisms.
