  • Bacteriophages originally isolated on a Gordonia terrae host were studied to determine their ability to infect three alternative Gordonia host species. Using spot testing and direct infection techniques, several sequenced G. terrae bacteriophages were assessed for their broad host range infectivity against G. rubripertincta, G. westfalica, and G. lacunae. Serial dilutions of high titer lysates were spotted on each host and efficiency of plate (EOPs) were calculated for each phage. While several phages provided positive spot tests on multiple hosts, their inability to produce direct infections on alternative hosts indicated that these results were the product of lysis from without. A mixed lysate of G. terrae phages Dogfish and Catfish provided a mid-range EOP spot test value, was able to directly infect G. rubripertincta, and was selected for further study to ascertain whether the observed broad host range infectivity was due to mutation or the presence of methylation. Our results indicate that several passages of G. terrae phage Dogfish on G. rubripertinta enhanced its infectivity on this alternative host, while repeated passages of Dogfish on its original host G. terrae did not. Sequencing of phage DNA isolated from Dogfish on G. rubripertincta indicated the lack of any mutation and points to the likelihood of methylation affecting host restriction enzyme systems on the alternative host.
