  • We analysed 222 blood samples (umbilical cord and/or peripheral) of healthy neonates, in order to obtain normal values for the lymphocyte population in neonates. A panel of combined monoclonal antibodies was utilized to obtain the maximal information in a minimal blood quantity. Excluding criteria were: mother with a chronic disease, signs of infection in the pregnancy or neonate, eclampsia, need of blood transfusion (ABO-incompatibility), medication with unknown side-effects, neonates with meconial waters, apgar index lower than 7 after 10 min., lactate $>$2.5 mmol/l, malformations in the neonate, time between sample collecting and cell marking over 12 hours, or coagles in the sample. After separating, the lymphocytes were washed and marked with the fluorescent monoclonal antibodies and resuspended in sheath fluid in order to be counted with the FACSCAN (Becton Dickinson). The antigen markers, their fluorescency and the calculated values (mean $\pm$ SEM) were as follows: CD1-FITC: 4.04%; CD2-FITC: 61.4%; CD3-PE:56.91%; CD4-FITC:35.18%; CD-PE: 23.55%; CD11b-PE: 24.50%; CD19-FITC: 11.98%; CD20-PE: 11.50%; CD25-FITC: 3.66%; TCR-FITC: 55.50%; TRR-FITC: 6.38%; HLADR-PE: 12.56%; IgG-FITC: 6.89%; Campath-FITC: 62.31%; Camplus-FITC: 62.84%; LFA-FITC: 57.26%. The results showed that T-lymphocytes in the neonate are lower than in later life ages. As lower the gestational age, the higher the immaturity of T-cells; the higher the gestational age, the higher is the maturity of B-lymphocytes. The interval between collecting and monoclonal marking of the cells plays an important role for the final results. An interval of longer than 12 hours leads to a decreased number of marked T-cells whereas the number of B-cells with antigenic markers is still growing.
