A paired domain and homeodomain‐containing transcription factor Pax6 is an evolutionarily conserved regulator implicated in the eye development of both vertebrates and invertebrates. Mutations of the Pax6 gene disrupt eye development in both mammals and insects while Pax6 misexpression can lead to the formation of ectopic eyes. Genetic and expression studies have indicated that the development of vertebrate eyes is guided by a conserved gene regulatory network with Pax6 being one of the key transcription factors. To define the regulatory inputs into Pax6 gene we performed a genetic dissection of its regulatory landscape. To uncover the direct target genes of Pax6 we performed chromatin immunoprecipitation (ChIP‐seq) using mouse embryonic retina as a sample. We will present our preliminary genetic data on functional validation of Pax6‐regulated enhancers.
