Evodiamine and rutaecarpin, major alkaloidal components of Evodiae Fructus (dried fruits of Evodia rutaecarpa var. bodinieri) were investigated for their antinociceptive and antiinflammatory effect. Oral administration of 30 or 100 mg/kg evodiamine significantly decreased the frequency of licking behavior in the late phase of the formalin test without affecting that of the early phase. Evodiamine inhibited not only the increases of vascular permeability induced by acetic acid and of paw edema induced by carrageenin, serotonin and bradykinin, but also the arachidonic acid-induced ear swelling. Rutaecarpine inhibited the effects of formalin test and the serotonin-, bradykinin- and arachidonic acid-induced edema. These results suggested that evodiamine and rutaecarpine possessed an antinociceptive effects mediated by antiinflammatory action, although the action mechanisms have not yet been clarified completely.
