Transthyretin (TTR) amyloid fibril formation is observed systemically in familial amyloid polyneuropathy and senile systemic amyloidosis and appears to be the causative agent in these diseases. Herein, we demonstrate conclusively that chromium(III) ion inhibits TTR fibril formation efficiently in vitro and in vivo, and does so by stabilizing the tetramer against dissociation and the subsequent conformational changes required for amyloid fibril formation. Further, chromium(III) ion and thyroxine (T4) at the physiological concentrations cooperatively inhibited amyloid formation. We also found the binding site of chromium(III) in the tetramer of TTR by examining the crystal structure. The sufficient intake of chromium(III) ion may be recommended to delay TTR amyloid fibril formation.
