  • Effects of various cations on the growth of tea and cucumber pollen tubes were studied to determine whether the pollen culture method could be employed for studying the mechanisms of salt stress and its alleviation in crops. NaCl at a concentration of 6-8 mM reduced the length of the tea pollen tubes to approximately 50% of the value of the control in the absence of divalent cations. The same degree of inhibition Was found with RbCI, LiCI, NH 4 Cl, and MgCl 2 . Stronger inhibition was observed with CsCI, BaCl 2 , and SrCl 2 , while a lower inhibition was recorded with KCl and CaCl 2 . Addition of 0.5-8 mM K or 0.5-4 mM Ca enabled to alleviate the inhibition induced by 6 mM NaCl. On the other hand, the length of the cucumber pollen tubes was reduced to approximately 50% of the value of the control at a concentration of 12-16 mM NaCl in the absence of Ca. The inhibition induced by 12 mM NaCl was alleviated by addition of 0.1-10 mM K or 4-10 mM Ca. The experimental results on pollen tubes were compared with the results reported in intact plants and the similarities and differences between them were discussed. The fundamental agreement in both experimental systems indicates that the pollen culture method can be used to study salt stress in crop plants.
