• This chapter discusses the combination of liquid chromatography and mass spectrometry (MS). MS is a powerful method for the identification of very small amounts of organic compounds and there is great interest in combining it with liquid chromatography (LC). MS serves as a sensitive method for the detection and identification of substances separated by LC. The combination of a mass spectrometer and a liquid chromatograph can be realized by either off-line or on-line coupling. An off-line combination is in use in most instances. The mobile phase is collected batch-wise and the fractions containing the substances of interest are then analyzed by mass spectrometry. Two main difficulties arise with the on-line coupling of a liquid chromatograph with a mass spectrometer. (a) In most mass spectrometers ionization of the molecules is achieved by electron impact ( E I ) or chemical ionization in the ion source. ( b ) The liquid stream leaving the chromatographic column cannot enter the mass spectrometer directly because of the high vacuum in the instrument.
