Epithelial to mesenchymal transition (EMT), an evolutionary conserved developmental program has been implicated in carcinogenesis, as EMT confers metastatic properties upon cancer cells by enhancing mobility, invasion, and resistance to apoptotic stimuli. Therefore, targeting EMT pathways constitute an attractive strategy for cancer treatment. However, challenges in accurately tracking dynamic epithelial-mesenchymal transitions as a function of the metastatic cascade has yielded conflicting results, and therefore determining the functional role of EMT in metastasis remains an area of active investigation. This chapter will focus on EMT in cancer progression, with emphasis on bone metastases, and discusses opportunities for novel anti-metastatic approaches for cancer therapy.
