The performance of any chromatographic system is based on the separation between adjacent sample components, and the spreading (dilution) of each component as it passes through the column. In elution chromatography, both of these characteristics are affected by the strength of the eluent and, hence, depend on the properties of the gradient in gradient-elution chromatography. To elucidate the effect of the gradient in ion exchange chromatography, a method for numerically solving a mathematical model of gradient-elution chromatography of multicomponent mixtures was developed which permitted a detailed study of the effects of varying the gradient. The model was based on the overall material balance and assumed first-order transfer rates between phases for each component. The effects of gradient changes and other parameters on elution volume and resolution could be studied independently by using this model whereas it would be very difficult and time consuming to do this experimentally. Examples of model-predicted chromatograms for various parameter changes are shown. A simplified model that predicts only elution position is also presented.
