Lohse, Roland Jahns Activating autoantibodies targeting the β1 -adrenoceptor may play a pathophysiological role in heart failure, but there is no reliable method for their detection. Here, we present a new method based on cyclic adenosine monophosphate (cAMP) measurements using a novel fluorescence sensor (Epac1-camps). Significant cAMP signals were induced by approximately 73% of immunoglobulin G fractions prepared from 55 dilated cardiomyopathy patients, 33% of them representing “high” activators (blocked by second extracellular β1 -receptor loop peptides), the other 40% “low” activators (blocked by first extracellular β1 -receptor loop peptides). In ischemic cardiomyopathy patients, we detected 5 high but no low activators. The immunoglobulin G prepared from 50 healthy subjects produced no cAMP signals. Our method facilitates rapid detection of different β1 -autoantibody classes and evaluation of novel therapeutic strategies.
