Intracellular impalements of oat coleoptile cells showing the highest input resistances (R) are always associated with less negative membrane voltages (V), whereas impalements with lower resistances display more negative voltages. Both fusicoccin (FC) and high external pH cause oat cells to hyperpolarize as much as 45 mV without altering input resistance. The characteristics relationship between R and membrane potential is therefore not the result of a heterogeneous population of cells displaying varying degrees of electrogenic proton pumping but reflects instead, as previously proposed, cells with two compartments, with the higher resistance barrier around the vacuole. The hyperpolarizing effects of FC and high external pH occur at the plasma membrane.
