• A method for the quantification of pregnenolone is reported. In this method the steroid is purified by paper and thin-layer chromatography and final quantification is by electron capture detection of 20β-hydroxy-pregnenolone-3,20-chloroacetate following gas chromatography. The method can be used for the accurate estimation of pregnenolone in biological samples when present in concentrations higher than 50 nanograms.
